260 Detection of CCND1 amplification in early lesions of nail apparatus melanoma using digital-droplet PCR

Backgrounds: Genetic alterations are strongly involved in carcinogenesis and pathogenesis of melanoma. Although non-acral cutaneous melanoma has been fully analyzed, acral mucosal need more investigation. Somatic copy number variation (CNV) is one the major genetic melanomas. Previous studies have suggested that CNV due to instability caused early palmoplantar melanoma, however it not yet evaluated lesion nail apparatus (NAM). We demonstrate detection from lesions NAM using ddPCR. Materials Methods: Six tissues were dissected out formaldehyde-fixed paraffin-embedded sections by laser microdissection.In situ lesions, invasive lymph node metastasis, if applicable, analyzed respectively. Copy numbers (CNs) ddPCR two probes designed for CCND1. Normal epidermis periphery each specimen was used as a normal control. Results: Three six including lesion, had CN amplifications CCND1 One nodular showed high amplification but lesion. The other case primary site metastasis. Conclusions: In this study, we occurred NAM, suggesting might be important well may useful lesions.
